Abstract

CYTOTOXIC AND NEPHROPROTECTIVE POTENTIAL OF A PROPRIETARY POLYHERBAL FORMULATION URIZONE – AN IN VITRO STUDY

ABSTRACT

There is an increasing trend in the incidence of renal disorders and this is mainly caused by exposure to chemicals or drugs like amino glycosides, acetaminophen, cisplatin, etc. In this present study, an attempt has been made to evaluate the cytotoxicity and nephroprotective potential of the hydroalcoholic extract of the proprietary polyherbal formulation ‘Urizone’ (HAEU) using cell lines. The cytotoxicity was evaluated using normal liver cells (Chang liver cell lines) and normal kidney cells (Vero cell lines) and the nephroprotective activity was evaluated against gentamicin induced cytotoxicity in vero cell lines. The cytotoxic and nephroprotective activity of the HAEU were evaluated by using MTT assay. The selected formulation does not produce any significant cytotoxicity in Chang and Vero cell lines at the concentration of 1000μg/ml and shows 65.9% and 69.86% cell viability respectively and the LC50 values were found to be 2516.12μg/ml and 4242.52μg/ml respectively. In the cytoprotective activity, gentamicin (50mM) treated cells showed 13.24% of viability. The HAEU at the concentration of 1000μg/ml showed an improvement in cell viability and it was found to be 30.46%. Hence, we conclude that, the formulation does not produce any significant toxicity in normal liver and kidney cell lines and it also reverses the toxicity induced by Gentamicin.
