Abstract

ROLE OF NON-INVASIVE METABOLOMICS AND GENOMIC MARKERS CIRCULATING MICRORNAS (miRNAs) FOR THE DIAGNOSIS AND PROGNOSIS OF DIFFERENT STAGES OF ENDOMETRIOSIS

ABSTRACT

Objective: The role of non-invasive metabolomic and genomic markers for the diagnosis and prognosis of the different stages of endometriosis patients. Materials and Methods: A total of 100 clinically confirmed female cases of endometriosis were enrolled in this study. Among 100 cases, 46 had minimal/mild (I/II) grade and 54 cases had moderate/severe (III/IV) endometriosis. 56 age and gender-matched control subjects were enrolled for comparative analysis. Both the tissue and peripheral blood samples were collected from all the participants. Serum was separated from fresh blood samples and miRNA was isolated from each serum sample within four hours of sample collection. Six different miRNA were selected for the present study and expression of each miRNA was measured using the relative quantification method based on SYBR-Green assay. Results: Expression of miRNA-21, miRNA-143, miRNA-146, and miRNA-155 was found to be significantly high in patients with endometriosis. Expression was high in stage III and IV compared to stage I and II. miRNA-141 and miRNA-200 have shown significantly reduced expression levels in endometriosis patients compared to controls. Expression was minimal in stage III and stage IV. More importantly, no significant difference was observed in expression levels of each miRNA between tissue and blood samples (p>0.05). Conclusion: The Panel of six-miRNA may be used as non-invasive metabolomic and genomic markers for the diagnosis and prognostics of the different stages of endometriosis.
