A REVIEW ON ORAL DRUG DELIVERY FOR THE APPLICATION OF COCRYSTALS

ABSTRACT
Among various routes, oral delivery has been recognized as the most
attractive method, mainly due to its potential for solid formulations
with long shelf life, sustained delivery, ease of administration and
intensified immune response. In oral drug delivery, many scientific
challenges and breakthrough technologies are required to generate
novel dosage forms raising drug delivery to higher level. The field of
pharmaceutical cocrystals has reached a tipping point, particularly
because cocrystals can improve the physicochemical properties of
drugs without compromising their therapeutic benefits. Poor aqueous
solubility and low oral bioavailability of an active pharmaceutical ingredient are the major
constraints during the development of new product. Cocrystals are homogenous (singlephase)
crystalline structures composed by two or more components in a definite
stoichiometric ratio bonded together by non-covalent bonds. Currently, cocrystals provide
exciting opportunities in the pharmaceutical industry for the development and manufacturing
of new medicines by improving poor physical properties of Active Pharmaceutical
Ingredients (APIs) such as processability, solubility, stability and bioavailability.
