Abstract

HIGH PERFORMANCE THIN LAYER CHROMATOGRAPHIC DETERMINATION OF PHYTOCONSTITUENTS IN HYDROALCOHOLIC AND METHANOLIC WHOLE PLANT EXTRACT OF Peristylus goodyeroides (D.DON) LINDL

ABSTRACT

Introduction: In Indian traditional healing medicinal orchid Peristylus Goodyeroides (D.Don) Lindl. based herbal formulation are used for gynaecological disease by the traditional healers of different regions. TLC is one of the best analytical methods for the separation and identification of phytoconstituents from the crude extract. The UV fluorescence method is employed for the detection of separated spots on the TLC Plates at 365nm. HPTLC is one of the sophisticated instrumental techniques for qualitative and quantitative analysis of the herbs and herbal drugs. It is modified version of TLC where separation of sample components is done on high performance layers with detection. Aim and Objective: The aim was to reveal the phytoconstituents from the selected herb using chromatographic detection. Method: The TLC parameters were set by trial and error method and the suitable mobile phases for the individual constituents. The phytoconstituents from both hydroalcoholic and methanolic extracts were identified at 365nm by HPTLC. Results and Discussion: The metabolic products of the plant were separated using respective mobile phases through Rf value. From the analysis, primary metabolites were found to be amino acids and secondary metabolites such as Alkaloids, Glycosides, Flavonoids Steroids and triterpenoids. Among these, only lipophillic compounds were choosed for HPTLC detection based on their therapeutic applications. Conclusion: The mentioned HPTLC profile for the plant of interest will hopefully help the researchers in herbal field to develop more herbal medicines requiring the particular constituent or its extract.
