PRODUCTION AND OPTIMIZATION OF AMYLASE FROM THERMOPHILIC BACTERIUM RHEINHEIMERA SP. DUPSA-2

Abstract In the present study, seven bacterial strains (designated DUPSA-1, DUPSA-2, DUPSA-3, DUPSA-4, DUPSA-5, DUPSA-6, and DUPSA- 7) were isolated from the Bendre Theertha hot spring water sample. Strains were evaluated for amylase activity using the plate assay method. Among the isolated strains, DUPSA-2 showed the highest amylase activity and hence was selected for further studies. Morphological, microscopic, and biochemical chemical characteristics preliminary indicate strain DUPSA-2 as a member of the genus Rheinheimera. The 16S rRNA gene sequence analysis showed it shared the highest similarity with Rheinheimera mesophile. Rheinheimera sp. DUPSA-2 showed optimum amylase activity at a temperature of 60°C, and pH 7.0 using 1% potato floor. The production of amylase at high temperature indicates its use for industrial application and hence further application will be carried out in our future studies.
