DNA NORMALITY AFTER IN VITRO PREPARATION WITH PENTOXIFULLINE AND GLYCYRRHIZA GLABRA OF CRYOPRESRVED HUMAN SPERMS

Abstract Many studies conducted on the pentoxifylline (PX) and Glycyrrhiza glabra (G.glabra) as motility stimulants showed positive effect on the activation of sperm in vitro and improve the forward movement. However, it was not known the impact of these stimulants on the nature of genetic material, especially after-cryopreservation of activated sperms in vitro. This experiment was designed to found out any harmful effect of medium contains a mixture of PX and aqueous extract of G.glabra on the sperm genetic material DNA after cryopreservation. One hundred men (normozoospermic and oligoasthenozoospermic patients), were involved in this current study. Ejaculated sperms were obtained from the patients and in vitro activation technique after cryopreservation-thawing was done by using two media namely; a mixture of PX with G.glabra (treated medium) and Ham’s F-12 alone (control medium) were performed to evaluate the certain sperm function characteristics as well as the evaluation of DNA normality by acridine orange test before and after cryopreservation and after activation of cryopreserved semen. The results of this study found that after cryopreservation of ejaculated human semen and in vitro preparation with a mixture of PX with G.glabra medium has shown a positive effect on sperm motility, grade activity of progressive forward movement and morphologically normal sperm (MNS) in both normozoospermic and oligoasthenozoospermic men compared to after thawing and control medium.
