SOLUBILITY ENHANCEMENT STUDY OF POORLY ABSORBABLE DRUGS

Abstract The solubility behavior of drugs is one of the most challenging aspects in formulation development. Most of the newly invented or naturally occurring phytochemical entities are poorly water soluble. As a result formulating them as oral solid dosage forms is a major hurdle to the specialists. Thus a greater understanding of dissolution & absorption behavior of drug with low aqueous solubility is required to successfully formulate them into more soluble and hence bioavailable drug product. Therefore different approaches are being explored to enhance the solubility of poorly water soluble drugs; one of such approach is using Solid dispersion technique . In this study, we are trying to increase the solubility of two synthetic drugs i.e. Meloxicam [chemically designated as 4-hydroxy-2-methyl-N-(5-methyl-2- thiazolyl)-2H-1,2-benzothiazine-3-carboxamide-1,1-dioxide] is a member of the enolic acid group of nonsteroidal anti-inflammatory drugs (NSAIDs) and hydrochlorothiazide (6-chloro-1,1-dioxo-3,4-dihydro-2H-1,2,4-benzothiadiazine-7- sulfonamide) which is diuretic and used in the management of hypertension. According to BCS classification meloxicam and hydrochlorothiazide is a class four drug having low aqueous solubility and low permeability. Major reasons contributing to the low plasma and tissue levels of meloxicam and hydrochlorothiazide appear to be due to poor absorption, rapid metabolism, and rapid systemic elimination. For this solvent evaporation and spray drying technique has been applied to improve the solubility and thus dissolution of poorly soluble and absorbable class IV drugs.
