FORMULATION AND EVALUATION SUSTAINED RELEASES PELLETS OF VENLAFAXINE HYDROCHLORIDE

Abstract
Depression is a state of mental illness. Depression can change a
person’s thinking/feelings and also affects his/her social behavior and
sense of physical well-being. It can run in families and usually starts
between the ages of 15 and 30 years. The purpose of present study to
formulate sustain release pellets of Venlafaxine HCl for sustain release
of drug to maintain the therapeutic window. The formulation process
was carried out in FBP by wurster technique. The work was performed
to extend/prolong the release of Venlafaxine hydrochloride by using
different polymers such as EC, HPMC. Our research includes
Preformulation study of drug and excipients, formulation and
evaluation, release kinetics and stability studies of pellets.
Preformulation studies were conducted on the drug and excipients used in the formulations
were found to be compatible. Venlafaxine pellets prepared by solution/suspension layering
technique using the mixture of polymers such as EC 10 cps and HPMC was found to be
relatively more efficient. These capsules can be used as such for oral sustain delivery of
Venlafaxine HCl up to 20 hrs. The prepared pellets were evaluated for various micromeritics
parameters and dissolution studies. The formulated and optimized formulation pellets of
Venlafaxine HCl were filled in a capsule for patient compliances. The Oral capsule of
Venlafaxine HCl. The capsule also evaluated for pharmacopeial parameters. The results of pellets were satisfactory and useful to treat patient of depression round the clock without swinging of mood.
