FORMULATION AND EVALUATION OF FAST DISSOLVING MOUTH FILM OF DOMPERDONE

. Abstract Fast dissolving oral films can be defined as a dosage form, which when placed in the oral cavity it will rapidly disintegrate and dissolves to release the medication for oral mucosal absorption. Domperidone consists antiemetic properties related to its dopamine receptor blocking activity at both the chemoreceptor trigger zone and at the gastric level. Domperidone is a prokinetic & works on the region in the brain that controls vomiting. It also acts on the upper digestive tract to increase the movement of the stomach and intestines, allowing food to move more easily through the stomach. The natural polymers like Pectin, Pullulan, and Karaya Gum were used as carrier in the formulation. Nine different formulations were prepared with varying ratios of polymer and excipients. The selections of natural polymers were based on their appearance, film forming capacity and folding endurance properties. It was found that, the prepared oral film of Domperidone that contains Pullulan showed the fastest disintegration time (61sec) among the other investigated polymers. The drug release rates were also observed, F1 formulation showed (Pullulan) 99.36 % drug release within 2 minutes with satisfactory mechanical properties. The overall results suggested that the prepared Domperidone fast dissolving oral films can be used as an attractive and alternative to the commercial available immediate release and conventional tablets resulting improved patient adherence.
