ASSESSMENT OF BARLERIA PRIONITIS STEM ETHANOL EXTRACT POTENTIAL TO KILL PATHOGENIC MICROBIAL STRAINS AND IN-VITRO INHIBITORY EFFECT ON HUMAN CANCER CELL LINES

Abstract
Objective: This study was designed to analyze the quality of Barleria
prionitis stems with ethanol extract for their evaluation of the potential
to inhibit the human cancer cells as well as pathogenic bacterial and
fungal strains. Methods: Extraction of Barleria prionitis stems in
ethanol was done by Soxhlet method and only seven secondary
metabolites were selected for further examination. After extraction,
phytochemical estimation of these selected seven secondary
metabolites i.e. alkaloids, flavonoids, anthroquinone, saponins,
terpenoids, tannins and cardiac glycosides was done as per the
protocols of Kokate. Minimum Inhibitory Concentration (MIC) effect
of Barleria stem ethanol (BSE) was done by using dilution method and
gradient plate methods on five bacterial and five fungal strains. Total activity of metabolites
was calculated through MIC. Whereas anticancer activity was done with the help of SRB
(Sulphorhodamine B) assay. Statistical analysis was done by using IBM SPSS statistics 20.0
and MS Excel 2007 with range (±standard deviation). Results: BSE exract with ethanol is a
non-polar solvent extract and considered as the best solvent to extract the maximum number
of secondary metabolites like alkaloids, saponins, tannins and flavanoids. BSE with 1.25mg
MIC, shown high antimicrobial activity against Pseudomonas aeruginosa, Chlamydia
pneumonia and Bacillus thruengiensis, the OD value of samples was 0.02±0.0005 (with
range 0.0211-0.0245),0.0309±0.0003 (with range 0.0311-0.0329) and 0.03±0.004 (with range
0.0301-0.0328) respectively. BSE with 6.25mg MIC, also shown high antifungal activity
against Cryptococcus neoformans, & Bacillus dermatitidis, the OD value of samples was 0.03±0.0003 (with range 0.0309-0.0329) & 0.03±0.0002 (with range 0.0310-0.0322) and showed moderate effects against C. Vaginitis, A. fumigates and A. niger. BSE extracts has found highest total activity against all 10 selected experimental strains (5-bacterial and 5-fungal strains).
