FORMULATION AND EVALUATION OF FAST DISSOLVING WAFER CONTAINING DOLUXETINE

. Abstract The rapid drug dissolution system offers a solution for patients who have difficulty swallowing tablets or capsules, etc. The wafer dissolves quickly when placed in mouth without need of water thus helpful in providing rapid onset of action. Doluxetine is a serotonin reuptake inhibitor (SRI) used as anxity and depression and depressant drug with extensive first pass metabolism which results in bioavailability. The present study was aimed to formulate fast dissolving wafer of doluxetine to enhance bioavailability and avoid pre-systemic metabolism to improve patient compliance. The fast-dissolving strips were prepared by solvent casting technique with the help of HPMC E15 and sodium starch glycolate(SSG). The strips were evaluated for wafer thickness, tensile strength, folding endurance, in vitro disintegration time, surface pH, drug content uniformity and in vitro dissolution studies. Official criteria for evaluation parameters were met for all formulations. Based on the evaluation parameters, the formulation F5 showed optimum performance against other formulations; hence formulation F5 was selected as optimized formulation.
