Abstract

FORMULATION AND EVALUATION OF ACHYRANTHES ASPERA LEAVES BUCCAL PATCHES FOR TOOTHACHE

Abstract

Toothache is a prevalent oral condition affecting a significant portion of the global population. The search for effective and convenient methods to alleviate toothache has led to the exploration of novel drug delivery systems. This study focuses on the formulation and evaluation of buccal patches utilizing Achyranthes spera leaves for the treatment of toothache. Buccal patches offer several advantages, including noninvasive administration, prolonged drug release, and localized therapy. The research objectives were twofold, first to develop a buccal patch formulation incorporating Achyranthes Aspera leaves extract, and second, to evaluate the patch's physicochemical properties and in-vitro drug release characteristics. The fabrication of the buccal patches involved selecting suitable polymers, plasticizers, and permeation enhancers to ensure optimal drug delivery. Achyranthes aspera leaves were chosen due to their well-documented analgesic and antiinflammatory properties, making them a promising natural alternative for toothache management. The formulation process involved solvent casting, where different combinations of polymers, such as hydroxypropyl methylcellulose (HPMC), ethyl cellulose (EC), and polyvinyl alcohol (PVA), were used to optimize the patch properties. Various plasticizers, including glycerine and propylene glycol, were incorporated to enhance the flexibility and adhesion of the patches. Furthermore, permeation enhancers such as menthol and eucalyptus oil were evaluated to improve drug permeation through the buccal mucosa. The buccal patches demonstrated promising physicochemical properties and sustained drug release behaviour. The use of natural ingredients such as Achyranthes aspera leaves may provide a safer and more accessible alternative for toothache relief.
