METAL-ORGANIC FRAMEWORK AN EMERGING CONCEPT IN DRUG DELIVERY SYSTEMS: NANOCARRIERS

. Abstract Metal-organic frameworks (MOFs) are a novel class of highly ordered crystalline porous coordination polymers that have potential applications in drug delivery systems. These frameworks made up of metal ions or clusters coupled to organic ligands, provide remarkable chemical functionality, tuneable porosity, and structural designability at the molecular level. This review looks at several synthesis methods, such as solvothermal, microwave-assisted, sonochemical, electrochemical, and mechanochemical approaches, and how they affect the efficacy of MOFs as nanocarriers in drug administration. The biomedical applications of MOFs, particularly in cancer treatment, are thoroughly examined. The encapsulation and regulated release of medicinal drugs like doxorubicin, cisplatin, and curcumin within MOFs represents a considerable improvement over traditional drug delivery system. MOFs' high loading capacities, stability, and ability to bypass physiological barriers improve efficacy and reduce systemic side effects. Additionally, the review covers the role of MOFs in wound healing, emphasizing their antibacterial and anti-inflammatory properties, and their potential in biosensors for early disease detection. The integration of MOFs with photosensitizers for photodynamic therapy and their application in oral delivery of protein therapeutics further underscore their versatility and potential in modern medicine. In conclusion, MOFs offer a promising platform for developing advanced drug delivery systems, with ongoing research to overcome existing limitations and explore new therapeutic applications.
