Abstract

EXTRACTION OF RUTIN FROM PHYSALIS PERUVIANA PLANT EXTRACT, ITS PHYTOCHEMICAL, ANTIOXIDANT, ANTIDIABETIC AND TIME KILL ASSAY

Abstract

The present study investigates the phytochemical composition, antioxidant, and antidiabetic of Physalis peruviana extract obtained from samples collected in the Nilgiris (Nilgiris, TamilNadu, India) region. The extract was prepared using n-hexane, followed by phytochemical screening, thin-layer chromatography (TLC), and column chromatography. The extract tested positive for terpenoids, flavonoids, and steroids, whereas alkaloids, phenols, sugars, saponins, quinine, and proteins were absent. TLC analysis revealed Rf values comparable to rutin (0.74), indicating the presence of bioactive compounds. The antioxidant assays demonstrated that the columnpurified product exhibited higher superoxide dismutase (SOD) (61.27 μg) and hydrogen peroxide (H2O2) scavenging activity (71.35 μg) compared to rutin (48.291 μg and 30.57 μg, respectively). The antidiabetic potential was evaluated through α-amylase and α- glucosidase inhibition, where rutin showed higher α-glucosidase inhibition (63.72%) than the column-purified product (49.75%), while the latter exhibited higher α-amylase inhibition (46.75%) compared to rutin (0.148%). The time-kill assay revealed dose-dependent cytotoxic activity, with rutin demonstrating a higher percentage of cell death at higher concentrations. These findings suggest that Physalis peruviana extracts possess significant antioxidant and antidiabetic properties, highlighting their potential therapeutic applications.
