ANTICANCER ACTIVITY TEST OF ETHYL ACETATE FRACTION OF MAJA FRUIT (AEGLE MARMELOS) AGAINST HELA AND MCF-7 CELLS

. Abstract Cancer is the leading cause of death in the world due to the malignancy of uncontrolled cell growth that damages other normal cells in sufferers. Genetic disorders, lifestyle, and environmental factors are the main factors causing the emergence of cancer cells in the body. The cause of this disease is the uncontrolled proliferation of cancer cells due to disruption of p21 function. One of the therapies to treat cancer that is widely used today is chemotherapy. However, this therapy produces side effects that can kill stem cells due to low selectivity. The discovery of anticancer drugs from natural materials continues to develop, so it requires an approach that provides an explanation of the potency of bioactive compounds in plants for medicine. Aegle marmelos is a plant that is potentially an anticancer agent because it has a variety of pharmacological activities. This study focused on the separation of the ethyl acetate fraction of maja fruit (Aegle marmelos) from its extract for anticancer activity testing using HeLa and MCF-7 cells. Extraction of maja fruit using maceration method with methanol solvent. Drying of extract using rotary vacuum evaporator until thick extract is obtained. The methanol extract obtained is then extracted using partition method with n-hexane, ethyl acetate and methanol solvents. The ethyl acetate extract obtained is dried using rotary vacuum evaporator until thick extract is obtained. Separation using gravity column chromatography method with n-hexane and ethyl acetate eluent system with a ratio of 8:2. The ethyl acetate fraction resulting from separation using the gravity column chromatography method was tested for anticancer activity with HeLa and MCF-7 cells. The IC50 value obtained from the ethyl acetate fraction of maja fruit (Aegle marmelos) was 212.50 μg/mL in HeLa cells and 398.11 μg/mL in MCF-7 cells.
