Abstract Inflammation is a fundamental biological response to injury and infection, but its dysregulation can lead to chronic conditions such as arthritis, diabetes, and cardiovascular diseases. The search for natural anti-inflammatory agents has gained momentum due to the adverse effects of synthetic drugs. Ziziphus jujuba (Jujube), a medicinal plant from the Rhamnaceae family, has been traditionally used for its therapeutic properties, including anti-inflammatory activity. This review explores the in-vitro anti-inflammatory potential of Ziziphus jujuba root extract, focusing on its phytochemical constituents, mechanisms of action, and potential medical applications. The root extract is rich in bioactive compounds such as flavonoids (quercetin, kaempferol), saponins, alkaloids, and phenolic compounds, which exhibit significant anti-inflammatory effects. Various in-vitro assays, including protein denaturation, membrane stabilization, nitric oxide (NO) inhibition, and cyclooxygenase (COX) and lipoxygenase (LOX) inhibition, demonstrate its ability to modulate key inflammatory pathways. Studies have shown that flavonoids in the extract inhibit COX-2 activity, comparable to ibuprofen, while saponins and alkaloids reduce oxidative stress and inflammatory mediator release. These findings suggest that Ziziphus jujuba root extract holds promise for the development of natural anti-inflammatory drugs, nutraceuticals, and cosmeceuticals. However, further in-vivo studies, pharmacokinetic analysis, and clinical trials are necessary to validate its safety and efficacy. The promising in vitro results highlight the potential of Ziziphus jujuba root extract as a valuable natural alternative for inflammation management and therapeutic d rug development.
