FORMULATION, DEVELOPMENT AND OPTIMIZATION OF TRANSDERMAL PATCH OF SCOPOLAMINE

. Abstract Objective: This research aims to develop a transdermal adhesive matrix patch of Scopolamine using Sterculia urens, enhancing patient compliance, minimizing side effects, and reducing dosing frequency for improved therapeutic outcomes. Methods: Transdermal patches were prepared using the solvent evaporation casting technique, incorporating Sterculia urens and sodium alginate polymers with a suitable plasticizer. The patches underwent physicochemical evaluation, including assessments of drug content, thickness, weight variation, and in vitro drug release using a Franz diffusion cell. Stability studies were conducted to optimize the transdermal patch formulation. Result: The prepared transdermal patches exhibited desirable physicochemical properties. The pH range (6.7-7.1) was compatible with skin pH, indicating minimal irritation risk. Thickness (0.028-0.036 mm) and weight uniformity (0.036-0.053 gm) were within acceptable limits. Folding endurance tests revealed optimal flexibility. Moisture uptake and content varied across batches (F1-F5), with batches other than F4 showing minimal moisture sensitivity. Swellability ranged from 63.15% to 78.26%, indicating good patch integrity. Batch F5 demonstrated the highest tensile strength (0.042 Kg/mm²). In vitro drug release studies showed >97% release across all batches after 480 minutes, with batch F3 exhibiting the highest release (98.82%) and F5 the lowest (97.33%). Notably, >60% of the drug was released within 240 minutes in all batches. Conclusion: This research concludes that scopolamine can be effectively formulated into adhesive transdermal patches, enabling sustained drug release over several hours.
