THE EFFICACY AND SAFETY OF FIXED DOSE SUSTAINED - RELEASE PREGABALIN, ACECLOFENIC AND METHYLCOBALAMIN IN SCIATICA AND PERIPHERAL NEUROPATHIC PAIN.

Abstract Objectives: To study the fixed dose combination of sustained release pregabalin, aceclofenic and methylcobalamine in neuropathic and sciatic pain and evaluate their efficacy, safety and tolerability. Materials and Methods: we conducted 14 day clinical trail and 138 patients were enrolled and conducted multi center non interventional study. patients were given fix dose of sustained-release pregabalin 75mg, aceclofenic 100 mg and methylcobalamin 500mcg for 14 days. During the 14 day time period patients were checked for side effects and also for symptoms of sciatic and peripheral neuropathic pain. Data collected for positive and negative symptoms such as burning, paresthesia, numbness back pain etc .we used ten-point visual analog scale for measuring pain intensity. Data was analyzed using statistical methods. Results: During the 14-day study there was significant improvement in positive and negative symptoms. From first week there was significant reduction in VAS score and mean VAS score on 14th day was 80.03%. Few adverse effects noticed like heart burn (2%), drowsiness (4.7%) and sedation (3.1%) but overall regimen was well tolerated by > 95% of patients. Conclusion: Fixed dose combination of sustainedrelease pregabalin, aceclofenic and methylcobalamin significantly reduces neuropathic and sciatic pain with improvement in Positive and a negative symptom associated with neuropathy and the regimen was well tolerated by patients.
