Abstract

FORMULATION AND EVALUATION OF EXTENDED RELEASE TABLET OF METOPROLOL SUCCINATE

Abstract

Extended drug delivery system deliver drug at a predetermined rate by maintaining a relatively constant, effective drug level in the body with concomitant minimization of undesirable side effects. This research work aim at formulation of extended release tablet with the help of Hydroxypropylmethylcellulose as matrix forming polymer in combination with ethyl cellulose by direct compression method. Metoprolol Succinate is a beta selective adrenoreceptor agent and is indicated for the treatment of Hypertension,Angina pectoris,Heart failure for the treatment of stable symptomatic heart failure of ischemic, Hypertensive.[3] Nine different formulations of Metoprolol Succinate Extended Release tablet were prepared. In each case the weight of the Metoprolol Succinate was kept constant i.e.25 mg. The ingredients were Metoprolol Succinate, matrix forming polymer Hydroxypropylmethylcellulose, Ethyl cellulose as a hydrophobic polymer and Magnesium stearate, microcrystalline cellulose as tableting aids. Prior to formulation preformulation studies were carried out. Tablets were prepared using direct compression technique and evaluated for various parameters like % drug content, Hardness, Thickness, Friability and In-vitro dissolution time. Erodible tablets were prepared using Hydroxypropylmethylcellulose polymer i.e. HPMC K4M. The erodible tablets were prepared in different concentration of Hydroxypropylmethylcellulose like 65mg, 70mg, 75mg, 80mg, 85mg, 90mg, 95mg, 100mg and 105mg. In vitro drug release study was carried out for tablets of all the formulations. The drug release kinetics carried out for all batches of the formulations. Studies were design to see the tableting properties and in vitro drug release profile of Metoprolol Succinate. From the result of the study of prepared tablets it is establishes that batch F8 containing Metoprolol Succinate 25mg Hydroxypropyl- Methylcellulose 105mg Ethyl cellulose 45mg Magnesium stearate 10mg Microcrystalline cellulose15mg was optimize batch. The release kinetics revealed that all the formulation follows zero order drug release with release exponent value (n) 0.7656.
