Abstract

PRODUCTION OF PHARMACEUTICALLY IMPORTANT BIOACTIVE MOLECULES BY FUNGAL ENDOPHYTES- SSD1 AND SSD2 FROM AXONOPUS COMPRESSUS

Abstract

Endophytes are symbiotic microorganisms present ubiquitously in plants which enhance ecological adaptability of host. Fungal endophytes are novel source of antimicrobial, insecticidal, anticancer and cytotoxic compounds. The aim of this research was to explore bioactive compounds from fungal endophytes from Axonopus compressus. Two fungal endophytes were isolated from the Axonopus compressus viz.SSD1 and SSD2. The fungal endophytes produced extracellular enzymes- pectin lyase, tyrosinase and asparaginase, metal nano particles- nickel and manganese, pigments and intracellular amino acids. Activity of pectin lyase for SSD1 and SSD2 were 2.07 U/ml and 1.03 U/ml respectively. Activity of tyrosinase for SSD1 and SSD2 were 4.90 U/ml and 8.05 U/ml, respectively. Activity of asparaginase for SSD1 and SSD2 were 4.20 U/ml and 4.93 U/ml, respectively. The approximate molecular weight (SDS-PAGE) of pectin lyase, tyrosinse & asparaginase were 39kDa, 130kDa, 138kDa for SSD1 and 34 kDa, 120kDa, 140kDa for SSD2, respectively. Nano particles produced from salt solutions of nickel and manganese by both the isolates show maximum absorbance at 400 nm. Two pigments, SSD1 - pinkish violet pigment and SSD2 orange pigment were extracted by Gadd method & its spectrophotometric analysis for SSD1 showed max at 240 nm & yield 7.78 mg/g and SSD2 showed max 230 nm & yield 9.52 mg/g. Rf values for intracellular amino acids - serine, tryptophan & isoleucine for SSD1 & SSD2 were 0.26, 0.64, 0.71 and 0.30, 0.60, 0.82, respectively. Therefore, bioactive products from fungal endophytes are gaining importance in agriculture, industry and especially pharmaceutical industry for their stability and environment-friendly technology.
