HPTLC AND RP-HPLC METHODS FOR THE ESTIMATION OF MANGIFERIN IN MANGIFERA INDICA EXTRACT AND ITS FORMULATION

Abstract
Two simple and accurate HPTLC and HPLC methods were developed
and validated for the estimation of mangiferin in Mangifera indica
(Anacardiaceae) and in its formulation using standard marker. Oxitard
capsule was selected for the current study. The validated HPTLC
analysis was carried out on plant extracts and formulation using ethyl
acetate : methanol : formic acid : water (10:1:1:0.5%v/v/v/v) as mobile
phase and the detection wavelength was 270nm. The Rf values of the
extract and formulation were compared with that of the standard. The
standardization of the extract and formulation were carried out by
validated HPLC method . The optimized solvent system was Methanol:
orthophosphoric acid (65:134%v/v). The flow rate was 1ml/minute. The detection
wavelength was 258nm.The plant extract was standardized, quantified and validated by
newly developed HPTLC and HPLC methods. The estimation of marker constituent present
in the extract and formulation can be utilized in routine estimation and standardization of
medicinal plant extract.
