A NOVEL APPROACH TO THE SYNTHESIS OF FLORFENICOL

Abstract
The article describes a convenient and ecofriendly synthetic route for
the synthesis of florfenicol from readily available thiamphenicol 1 by
using inexpensive and recyclable solid catalysts. The strategy is based
on the preparation of cyclized alcoholic oxazolidin-2-one 3. This
compound was protected with mesityl chloride in DCM and added Cu-
HAP as a catalyst than followed by fluorinated with KF/PEG and
added HAP as a catalyst to form fluorinated oxazolidin-2-one 5. This
compound was opened up by reaction with sulfuric acid in dioxane and
later the amino group was dichloroacetylated with
methyldichloroacetate by copper hydrotalcite (Cu-HT-2) in DCM to
obtained florfenicol 7 with good yield. All the synthesized compounds
were established by their spectral and elemental analysis.
